Unable to auto-open repository after installing latest update

I recently updated an installation of the Enterprise (64-bit) version to the latest version (v6.3.12.4, believe it was on v6.3.11.x prior), and now when I attempt to launch the application (which is set to auto-open the repository) it throws the attached error messages.

If I go into the Options... panel, I can perform a test connection without any issues. But attempting to reconnect through there (or the main menu) will trigger said attached errors.
